Can I buy a PSN Card from US region then used it here in Singapore assuming that my Media go is set to US also? I'm worry if the PSN will check my IP and ignore my purchase.

But you cannot switch the region of your PSP too often apparently, it may permanently refuse to activate after a couple of switches. So this procedure is a bit risky.

If my PSP is set to Singapore region and my Media go set to US region, then I successfully download the game to my Media Go, is there any problem if I transfered that game to my PSP?

I dont think they track ip's, I live in India and used a European as well as US account on psn, All demos worked fine
But you cant simultaneously use both accounts, so I stick with US. I bought my PSP in Netherlands and my country on my psp is India.

It might be because you try the demo game only. My problem is I try to buy the games. It wont allowed to add funds on my wallet.
Error: 80023102 or The funds could not add to your wallet (even I put the correct details).

You cannot add funds with a Non US card on the US PSN, I've had this problem several times, so I think that's your problem too: Sony North America doesn't want our dirty foreigner money

How about If I buy PSN Card from US then setup my Media Go to US region. Is that possible to accept my purchase even my current location is Singapore?

Yes that could work, assuming you can buy a PSN card from the US... (for example, not doable on Amazon with a foreign card AFAIK)
I'm also wondering if PSN is checking your default address...
